The promoter sequence is modified by disrupting both Fis binding sites where an inhibitory regulator binds; moreover,since there is an overlap between the activator and inhibitor binding site, we had to make sure the activator can still bind on the same site. The modified promoter is inserted in to pClone Red (J119137). The goal is that nitrate induces the promoter, then RFP is expressed when nitrate is present.
