Description

Inserting a NotI/EcoRI fragment into the PspOMI/MfeI sites of J61017 destroys the restriction sites and introduces standard Biobrick flanking sequence. In the process, the OnRFP cassette is destroyed allowing Red to White screening of colonies.

The registry sequence is blunt relative to XbaI/SpeI.
