Description

This generator is under control of extracellular glucose concentration and insulin activation state. High extracellular glucose concentration will turn off the generator by the binding of phosphorylated OmpR. Unless BBa_I761007 has respond to prior high extracellular glucose concentration, this generator will not turn on. This is achieved by the control of cinr and cinl. After mouse insulin degradation enzyme is generated, it will be exported and can degrade extracellular insulin.

Notes

This generator will turn on if and only if the extracellular glucose concentration is low and BBa_I761007 has been turned on at least once. The basic concept is that when extracellular glucose concentration is low, OmpR dephosphrylated and dissociate from its binding site. When this occurs, BBa_I761007 turns off, but the inhibitory effect of phosphorylated OmpR on this generator is relieved. While the mRNA synthesis of cinr and cinl has turned off, there are still some active CinR and CinL presented in E. coli as protein. Therefore, the generator will turn on due to the activation of the promotor BBa_R0078. Hence our goal is achieved.
