Sequences BBa_J100210_sequence (Version 1)

Description

Using Golden Gate Assembly, the transcriptional riboswitch RS4 (from Wachsmuth, et al.) was ligated into the destination vector tClone Red (J119361) in place of the insert (between BsaI sites). This riboswitch is controlled by theophylline, allowing transcription if theophylline is present. However, it also has shown increased transcription levels in the presence of caffeine.

Notes

Appropriate sticky ends had to be added to the RS4 sequence in order to anneal with those created by digestion of the destination vector with BsaI.

Source

The destination vector is part J119361. The sequence of the riboswitch comes from the paper "De novo design of a synthetic riboswitch that regulates transcription termination," by Wachsmuth, et al. (referred to as "RS4" in that paper; doi:10.1093/nar/gks1330).
