Description

Started with tCloneTet (J119367) and used Golden Gate Assembly to remove GFP reporter and put transcriptional terminator riboswitch RS10shift (RS10s). RS10s was taken from the Wachsmuth et. al. 2013 NAR vol. 41 p. 2541. This riboswitch works and is responsive to theophylline (2.5mM).

Notes

Riboswitch cannot have internal Bsa1 restriction site.
