Description

This construct is built by modifying the promoter sequence of Original Nitrate Biosensor (J100237), which inserts narG promoter sequence into pClone Red (J119137) through GGA. The promoter sequence is modified by disrupting the left Fis binding site, where an inhibitory regulator binds; moreover,since there is an overlap between the activator and inhibitor binding site, we had to make sure the activator can still bind on the same site. The modified promtoer is inserted in to pClone Red (J119137). Nitrate induces the promoter, so RFP is expressed when nitrate is present.

Notes

There is a deletion at BD18 bicistron RBS.
