Fusion protein of tetA and sacB connected with a linker sequence, with a pBAD promotor and RBS. TetA encodes tetracycline resistance and sacB kills E. coli in the presence of sucrose, allowing for both positive and negative selection. pBAD is an arabinose-inducible promoter.

Notes

Linker sequence was modified to optimize the GC content of the primers.
