Description

This part consists of the Toxin mazF which is cappable of activating cell death. This Biobrick includes an inducible promoter (pLac) enabling us to IPTG inducible kill Bacteria.
We tested this part with a weaker RBS (B0032) and a stronger RBS (B0034 this part). This part is cappable of inducing cell death in bacteria.

MazF is part of the biological toxin-antitoxin system mazEF. This system is derived from the bacterium Escherichia coli, where it serves as a stress inducable killswitch. mazF, a RNA-degrading enzyme, ist he toxin and is normally inhibited by the presence of mazE. Absence of mazE mazF leads to cell death.

Notes

We first constructed the part with two RBS of different strengths (B0034 and B0032). While the part with the weaker RBS (B0032) did not show the ability do inducible cell death this part (B0034) was cappable of IPTG inducible kill bacterial cells.
