Description

The plasmid contains the four necessary genes (dxs, crtE, crtI and crtB) for the biosynthesis of beta-carotene in an operon design. This means that you need just one promoter to express all four genes that are needed for the biosynthesi. The driving promoter is a pT5 that is used in our host lab. Furthermore, an origin of replication for E.coli is included. This origin of replication makes the plasmid high-copy. We decided for the BioBrick backbone with an Ampicillin resistance because we want to use it in the future in combination with another plasmid that carries Chloramphenicol resistance.

Notes

To construct this composite part, we used novel DNA assembly methods as CPEC or Gibson. This composite part is not BioBrick compatible because it contains BioBrick cut sites in the genes for beta-carotene biosynthesis.
