Description

We designed and built a kill switch mechanism that based on the activation of a lethal gene downstream of an inducible promoter that is activated over time.


The presented part is the first and the second module of our designed genetic circuit but without AiiA which consist of a constitutive promoter (I14032) driving expression of LuxR followed by a Lux promoter (R0062) , driving expression of TetR.


We built this BioBrick to test the degradation of AHL by AiiA and how AiiA affects the system.


You can use this BioBrick to produce TetR for a longer period compared to BioBrick BBa_K17600x (add the number of the full construct with AiiA) as it lack the AiiA, which is an enzyme that degrades AHL hence AHL degrades slower. . Hence the gene that will be cloned downstream of the TetR promoter will be expressed later in time.
