Description

This part produces a synthetic repressor in response to T7 RNA Polymerase. This synthetic repressor consists of a fusion of two DNA binding domains connected with a linker that is sensitive to matrix metalloprotease 9, and it represses the promoter described by parts BBa_K1833001 and BBa_K1833003. For more information on this part, visit 2015.igem.org/Team:Pitt/Protease/Project.

Notes

The part was synthesized de novo. For simple cloning, there are no smaller parts used to make this part.

Source

The two protein domains comprising the fusion protein was used as in "A two-hybrid system based on chimeric operator recognition for studying protein homo/heterodimerization in Escherichia coli" (G. Di Lallo, L. Castagnoli, P. Ghelardini and L. Paolozzi, Microbiology (2001), 147, 1651???1656). The linker sequence was used from "Fluorescence Detection of MMP-9. I. MMP-9 Selectively Cleaves Lys-Gly-Pro-Arg-Ser-Leu-Ser-Gly-Lys Peptide" (Rafal Fudala et al, Curr Pharm Biotechnol. 2011 May ; 12(5): 834???838.)
