Description

The stf gene encodes the short tail fibres found on the wild-type strain of bacteriophage lambda. The gene has been inactivated by a single frameshift mutation in the lab strain most commonly used in research (λ-PaPa). This gene has been created using the ORF-314 and ORF-401, cloning the two open reading frames into a single in-frame coding sequence through the use of an Eco-RV restriction site.

The C-terminal end of the stf protein offers host receptor recognition for OmpC on the surface of E.coli, widening the host range of λ-PaPa, which recognises only the lamB protein through its tip attachment protein. Additionally, this gene displays a high level (~40%) of homology with the gp37 of bacteriophage T4.

Notes

The gene was optimised to remove illegal restriction sites to make the sequence BioBrick-compatible.
