Description

This part contains just the coding sequence for the N-carbobenzyloxy (CBZ)-cleavage enzyme necessary to enantioselectively remove the CBZ protecting group from amino acids. In our biocontainment system, the enzyme serves to cleave CBZ from leucyl-tRNA charged with CBZ-protected leucine. This part can be used by other iGEM teams to selectively cleave the CBZ protecting group from amino or hydroxyl groups in organic reactions.

Notes

The DNA sequence was found in a study by Patel et al. In order to remove forbidden restriction sites from the coding sequence, silent mutations were made to remove the restriction site while preserving the amino acid sequence. The piece of DNA was synthesized commercially.
