Description

The complete coding sequence of Ovalbumin (OVA) acquired from GenBank (Accession# AH002466.2) . OVA has been commonly used as the antigen for testing the efficiency of antibody response and T cell activation in previous immunology experiments, thus is a easy to use antigen.

Notes

The Ovalbumin (OVA) sequence is a 2098 bp DNA sequence acquired from GenBank (Accession# AH002466.2). We changed several nucleotides of the sequence to avoid the restriction sites and separate the sequence into two parts since it???s too long to synthesis. The restriction site between these two parts is SapI. After cutting the sequence with EcoRI, SapI and PstI, we ligated the two parts with pSB1C3 and confirmed the OVA sequence is correct by DNA sequencing. In order to compare the protein expression efficiency of our expression vector and the original pX63HYG shuttle vector, we added BamHI restriction site at the head and tail of OVA coding sequence for the ligation of pX63HYG shuttle vector and OVA sequence.
