Description

This is a fosmid based on pCC2Fos from EPICENTRE Biotechnologies (http://www.epibio.com/item.asp?ID=385). It contains 38 kb from the human MECP2 genomic locus, with a spectinomycin resistance (SpecR) cassette in the first intron of MECP2.

This fosmid is used to accept by recombineering the HincII fragment from BBa_K260016 (inside backbone BBa_K260010). This reaction also transfers the blasticidin resistance gene (BsdR) to generate the new backbone BBa_K260001 (pCC2FosMB), then containing the same BBa_K260016 part.

Notes

Intron 1 of MECP2 previously contained a BsdR cassette. This had to be changed to SpecR by recombineering to generate the present fosmid.
