Description

This part is made of the lac promoter followed by 356K firefly luciferase mutant, which shifts its emission spectrum to make it red. Note that firefly luciferase is temperature sensitive, and cells grown at 30??C are considerably brighter than cells grown at 37??C.

The luciferase uses luciferin as a substrate, which needs to be added to the medium.

Notes

Note that the luciferase is temperature sensitive and cells grown at 30??C are a lot brighter that cells grown at 37??C.
Luciferin penetrates the cell much better at low pH. If the cells are spun down, then resuspended in citrate buffer, pH4.8, they are much brighter.
