Description

The biobrick contains the cellulose synthesis related genes situated at the upstream and downstream of the cellulose synthase operon, which have been proposed to increase the production of bacteria cellulose, altough their function has not been completely elucidated. This biobrick contains the cmcax and ccp genes in 2,092 bp containing SpeI and PstI restriction sites cloned in pSB1C3. The cmcax (endo-1,4-β-glucanase) is required to enhance the cellulose biosynthesis by providing carbon source to acs operon whereas the ccp encoding the cellulose complementing protein.

Notes

The cmcax gene has 2 Pst I cutting site while ccp gene has a EcoRI cutting site. During ligation, Spe and AlwN I is used to do enzyme digestion to get the cmcax sequence (approx. 1kb), followed by the binding of pSB1C3. Next, AlwNI and PstI will be used to do the second enzyme digestion in order to get ccp sequence (approx. 1kb), followed by the binding of pSB1C3. It is necessary to avoid restriction of wanted DNA while intentionally cutting the ends of DNA.
