Description

Blue Light Promoter (BLP) controlling Chloramphenicol Resistance (Chlor) on pSB1A3 vector

Reporter ycgF responds to blue light changing its conformation and dimerizing with ycgE molecules. Because of the dimerization, ycgF releases from the promoter region. Consequently, ycgF no longer represses gene transcription. Downstream, the gene for chloramphenicol resistance is now accessible to transcription enzymes. Thus, blue light initializes chloramphenicol resistance.

Notes

The use of the psB1A3 as opposed to the psB1C3 backbone:
The device produces chloramphenicol resistance in the cells. The backbone pSB1C3 has chloramphenicol resistance on its own and, if used as our backbone, would be redundant. It would not be possible to screen which cells were expressing the blue light promoter because chloramphenicol resistance would not be a guaranteed indicator of BLP expression.
