Description

The twin argenine tag (TAT) sequence targets proteins to the membrane for secretion. This signal sequence contains the conserved T-R-R-X-F-L-K sequence motif at the end terminus.

The red fluorescent protein is used as a proof of principle to confirm that the TAT signal sequence is capable of sequestering the protein outside of the cell.

Notes

Red fluorescent protein is an easily detectable reporter, allowing for easy proof of principle.

PCR was done using DNA primers designed for E1010, adding the TAT sequence to the 5' end. A VR primer was used for the 3' antisense.
