Sequences BBa_K743015_sequence (Version 1)

Description

This part is designed for the expression of the LuxA and LuxB genes from Vibrio fischeri in a circadian manner, peaking a 2 hours after subjective dusk (or hour 14 of circadian time if you prefer).
When transfromed into Syenchocystis PCC6803, the endogenous circadian transcriptional mechanism controls the activity of the Pta promoter making the heterodimeric LuxAB luciferase to be expressed at the specified time.
This permits for the emition of light by the luciferase reaction to be restricted to this time of the day (dependent on the addition or endogenous production of substrates)

Notes

This part was made by Gibson assembly, therefore it has no scars

Source

Pta promoter was amplified from genomic Synechocystis DNA, you can find a biobrick version of this promoter at blahblahblah.
LuxAB genes were obtained from the Luxbrick
