Description

The promoter is activated when: (1) there is no glucose in the medium and (2) there is lactose in the medium. Our construction uses the well-known lactose operon system. A lack of glucose promotes the formation of CAP (or CRP), which binds to specific sites upstream of sugar-metabolization genes and activates its transcription. Moreover, another condition is needed since LacI repressor will bind to the operator region and will block the transcription unless lactose is also in the medium.

The sequence of the promoter comes from E. coli genome, whereas the sequence encoding the AmCyan fluorescent protein is not natural, since it was optimized for codon usage and maximum fluorescence production.
