Description

This is a signalling system comprising two open reading frames of the ttrR and ttrS genes together with the promoter/operator of the ttrB gene from Salmonella LT2. Together, TtrR and TtrS form a canonical two-component system where TtrS is the membrane-bound sensor kinase and TtrR is the DNA-binding response regulator. The system senses tetrathionate outside the cell and activates transcription of the ttrB gene, which is part of a tetrathionate reducatase. This part has been designed so that expression of alternative reporter genes can be regulated by tetrathionate.

Notes

A PstI site in the natural ttrS sequence has been silently removed.
