Description

We have a high efficiency constitutive promoter and RBS preceding the signal sequence for maximum production. We used the PELB signal sequence from E. coli to guide the protein into the periplasmic space. We added a histidine tag to the end of the Oxytocin-NeurophysinI gene. This protein is also known as prepro-oxyphysin.

Notes

We used the PELB signal sequence from E. coli because the Oxytocin protein begins with a cysteine that forms a disulfide bond with the cysteine in position six. This cysteine must be free of any other amino acids. The PELB signal sequence guides the protein to the inner membrane of E. coli and the signal sequence is cleaved as the protein enters the periplasmic space, leaving the position one cysteine free to form disulfide bonds.
