Description

In their 2014 paper "Negative selection and stringency modulation in phage-assisted continuous evolution," Carlson et al., designed part J100265 to be used as an accessory plasmid in phage-assisted continuous evolution (PACE). This DNA part comes from a collection of 8 plasmids numbers J100264 - J100271. PACE employs M13 phage and E. coli in a chemostat. In order to conduct a PACE experiment of our own we used PCR and GGA to replace the LuxAB gene on part J100265 with the GFP gene to construct part J100287.

Notes

We sought to replace LuxAB with GFP because we have greater familiarity with GFP fluorescence than we do with LuxAB luminescence.
