Description

It encodes whole luciferase under the control of pBAD promoter. And we add arabinose until its final concentration is 1mmol/L, incubate for 6h(37??C, 250rpm). We can detect its activity by add its substrate fluorescein.

Notes

The DNA sequence of luciferase gene is from NCBI (Genebank:M65067.1). A pBAD promoter(BBa_K206000) and a RBS(BBa_B0034) are added upstream the functional gene to produce this enzyme. Lociferase-pSB1C3 plasmid is transformed into E. coli strain transB (DE3) to express the protein. It can show fluorescence when we add its substrate fluorescein.
