Description

Construct assembled to allow the quantitative analysis of the heat shock/LacI promoter/repressor system, BBa_K338002. Produces GFP in response to low-temperature heat shock, more-so in the presence of LacI. GFP production is retarded in LacIq cells (or in the presence of +5mM glucose) about 4-6x. See K338002 for more detailed data.

Notes

The type of GFP used in this construct happens to have a very long half life. If growing cultures for fluorescence measurements, grow at 30??C - any higher will activate the promoter, causing GFP to be produced.
