Description

Tuneable Temperature Sensor made with parts available in Registry 7.05 DNA-1. At higher temperatures, the lacI dimer degrades into lacI monomers. The LacI promoter is no longer inhibited, hence more RFP is produced. Also, cI 434 is produced resulting in Prm+ being repressed, thus lowering the levels of GFP. At lower temperatures, the lacI dimer is allowed to form, hence the LacI promoter is repressed. RFP levels decrease, and cI levels drop. This results in Prm+ no longer being repressed, hence GFP levels increase. Behaviour is tuneable by adjusting the levels of lacI ts via L-arabinose activating pBad. The net result is that the fluorescence (green to red) can be correlated to a temperature (at a given level of L-arabinose). Note IPTG induces R0010, offering a second degree of tuning.
