Description

OriT is the nic region on the F Plasmid that allows for conjugation in the presence of transfer genes encoded in TraJ genes. This OriT gene is followed by an autolysin N-acetylmuramoyl-L-alanine amidase. This codes for an Autolysin that will cause cell death to the cell upon conjugation. This composite part is meant to be incorporated into a plasmid with the TraJ genes knocked out with a kanamycin resistance gene. This will further prevent conjugation by the bacteria.

Notes

There may need to be up-regulation of the autolysin in this case to increase the rate of cell death.
